Experimental Verification for 1o7k » PX domain of NADH oxidase (p47phox), ligand-bound
This is membrane-association mode with specifically bound phosphoinositol lipid: the interaction occurs through W80, in agreement with experimental studies (Stahelin et al. 2003). Calculated hydrocarbon boundary of the lipid bilayer corresponds to the carbonyl groups of the bound lipid. Lipid-free structure of p47phox had two alterantive membrane-binding modes with nearly identical transfer energies: (1) through I60 and I65 and (2) through hydrophobic beta-turn F18-V19 (ligand-free p47pox associates with membrane through I65 according to Stahelin et al. 2003). The exposed hydrophobic areas are masked in the inactive state of the PX domain (Karathanassis et al. 2002). Transfer energy was calculated with bound lipid.
